Project: Next Generation Sequencing: From Computational Challenges to Biological Insight

Abstract: Molecular basis of chronic inflammation associated with obesity and its role in development of insulin resistance and atherosclerosis are poorly understood. In our project we use THP-1 macrophages under metabolic and inflammatory stress conditions to investigate transcription regulatory mechanisms characteristic for obesity-induced inflammation. Using single-cell RNA sequencing technology we will be able to systematically reveal regulatory networks and estimate heterogeneity in macrophage population. This application will enable us to discover diseased cell characteristics and to develop new approaches for prevention and treatment of age-related diseases such as diabetes.
